How to select a cell culture plate? 

1) The cell culture plates can be divided into flat bottom and round bottom (U-shaped and V-shaped) according to the shape of the bottom;
2) The number of culture holes was 6, 12, 24, 48, 96, 384, 1536, etc;
3) According to different materials, there are Terasaki plate and ordinary cell culture plate. The specific selection depends on the type of cultured cells, the required culture volume and different experimental purposes.

The difference and selection of flat and round bottom (U-shaped and V-shaped) culture plates

All types of flat bottom cells can be used, but when the number of cells is small, such as cloning, 96 well flat bottom plates are used.

 

In addition, when doing MTT and other experiments, the flat bottom plate is generally used for both adherent and suspended cells.

 

As for U-shaped or V-shaped plates, they are generally used in some special requirements. For example, in immunology, when two different lymphocytes are mixed, they need to contact each other to stimulate. Therefore, U-shaped plates are generally required. Because cells will gather in a small range due to the effect of gravity, V-shaped plates are less useful. V-shaped plates are usually used in cell killing experiments to make target cells closely contact, but U-shaped plates can also be used in this experiment (after adding cells, centrifuge at low speed).

 

If it is used for cell culture, it is usually flat bottomed. In addition, special attention should be paid to the material. The mark “Tissue Culture (TC) Treated” is used for cell culture.

 

The round bottom is usually used for analysis, chemical reaction, or sample preservation. Because round bottoms are better for absorbing liquids, and flat bottoms are not. However, if you want to measure the light absorption value, you must buy a flat bottom one.

 

Most cell cultures use flat bottom culture plates, which are easy to observe under the microscope, have a clear bottom area, have relatively consistent cell culture liquid level height, and also facilitate MTT detection.

 

The round bottom culture plate is mainly used for the experiment of isotope incorporation, and the cell collection instrument is required to collect the cell culture, such as “mixed lymphocyte culture”.
