Preparation before cell culture

Periodic inspection of cell culture

Regular examination of the morphology of cultured cells, that is, shape and appearance, is essential for the success of cell culture experiments.
In addition to confirming the healthy state of cells, examining cells with naked eyes and microscope each time you operate cells can also find signs of pollution early, so as to avoid the spread of pollution to other cells in the laboratory.
Signs of cell degeneration

The signs of cell degeneration include the appearance of granules around the nucleus, the dissociation of cells from the matrix, and the formation of cytoplasmic vacuoles.

These metamorphic signs may be caused by many reasons, such as:

Contamination of the culture, cell line senescence, or presence of toxic substances in the culture medium, or these signs only indicate that the culture needs to be replaced.
When the metamorphism is serious, it will become irreversible change.

Disinfection and layout of cell culture fume hood

Keep the cell culture fume hood clean and orderly, and place all objects within the direct view range.

Spray 70% ethanol on all articles put into the fume hood, wipe and clean them for disinfection.

Place a cell culture container in the open place in the middle of the fume hood; The pipette is placed at the right front for easy access; The reagent and culture medium are placed at the right back for easy absorption; The test tube rack is arranged at the middle rear part; A small container is placed at the left rear to hold waste liquid.
