A practical guide to tissue fixation, embedding and sectioning
Generally, it is necessary to first obtain material→fix→transparency→embedding→section, and finally carry out operations such as staining and histochemistry. This article focuses on how to embed and slice after the material is collected.
Embedding can be divided into OCT embedding (frozen section) and paraffin embedding.
1) Frozen sections are better for preserving the antigenicity of the tissue, but poor for morphological preservation, and are generally used for in situ hybridization (ISH) and immunofluorescence IF. Frozen sections are thicker and generally have a thickness of 8-15 μm. The frozen embedding step is relatively simple. After fixing, place the sample directly on the sample holder with tweezers, squeeze the OCT to completely wrap the tissue, and put it in the freezer until the OCT is solidified. Once solidified it can be sliced directly. If it cannot be sliced in time, wrap it in tin foil and store it at -80°C.
2) Paraffin sections are generally used for HE staining or immunohistochemical IHC. The thickness is thinner, generally 4um, the process is more complicated, and the quality of the sample depends on the type and composition of different tissues. It is more dependent on the experience of the producer, and it needs to be continuously optimized during the experiment to obtain the best experimental conditions.
