The answer is sometimes yes, sometimes no, depending on the type of preparation and the requirements of the microscope.
The most critical information can be found on the inscription on the objective lens. What do the bunch of things marked on the microscope objective lens mean?
1. The type of objective lens:
APO: Apochromat Objectives
FL: Fluorite objective lens
PL or Plan: Plan Objective
2. Magnification: This is generally the most conspicuous number. Most manufacturers will use color to distinguish objectives of different magnifications
3. Numerical aperture: usually 0.25, 0.65, 1.3, etc.
4. Standard mechanical barrel length: usually 160, 170 or ∞
5. There are three cases where cover glass is required:
The first type: if a cover glass is required, the thickness of the required cover glass will be indicated, such as 0.17.
The second type: No cover glass is required (and no cover glass is allowed), and a 0 or no mark will be marked
The third type: it doesn’t matter whether it is covered or not, it will be marked with “-”
6. Immersion system: if it is an oil lens, there will be the word “oil”
